[Pharmacological action] Pharmacodynamics: albendazole is a kind of benzimidazole, which has a broad-spectrum anthelmintic effect. Nematode is sensitive to it, and also has a strong effect on tapeworm and trematode (but needs a large dose), but has no effect on schistosome. The mechanism of action is mainly to combine with the tubulin of nematodes. After albendazole binds to tubulin, it prevents it from polymerizing with a-tubulin to form microtubules. Microtubules are the basic structural units of many organelles, which are necessary for cell reproduction processes such as mitosis, protein assembly and energy metabolism. The affinity of albendazole to nematode tubulin is significantly higher than that of mammalian tubulin, so albendazole has little toxicity to mammalian. This product not only has a strong effect on adults, but also has a strong effect on immature worms and larvae, as well as killing eggs.

 

Pharmacokinetics: Albendazole is a benzimidazole drug with good absorption after oral administration. Cattle can absorb 50% of the dose from the gastrointestinal tract. Within 9 days, 47% of the drug metabolites of oral dose can be recovered from urine. After oral administration in sheep, because it is metabolized to albendazole sulfoxide (the main active metabolite of anthelmintic effect) soon, the prototype drug can not be detected in the blood or can only be detected for a short time. At 20 hours after administration, the metabolites albendazole sulfoxide and albendazole sulfone reached the plasma drug peak concentration. The elimination half lives of sulfoxide metabolites in cattle, sheep, pigs, rabbits and chickens were 20.5, 7.7-9.0, 5.9, 4.1 and 4.3 hours respectively, and the elimination half lives of sulfoxide metabolites were 11.6, 11.8, 9.2, 9.6 and 2.5 hours respectively. In addition to sulfoxide and sulfone, there are hydroxylation, hydrolysis and binding products, which are discharged from the body through bile.

 

[Drug interaction] The combination of albendazole and praziquantel can increase the blood concentration of the former.

[Usage and dosage] Take orally: 0.2~0.4 tablets per 1kg body weight; 0.4~0.6 pieces of cattle and sheep; 0.2~0.4 pigs; 1~2 dogs; 0.4~0.8 pieces of poultry.

 

[Adverse reactions] (1) Dogs treated with 50mg/kg twice a day will gradually develop anorexia.

 

(2) Cats may have mild sleepiness, depression, anorexia and other symptoms. When using this product to treat paragonimiasis, there is a phenomenon of anti drug use.

 

(3) Can cause aplastic anemia in dogs and cats.
